Mikromorfologi dan Perkecambahan In Vitro Biji Anggrek Endemik Sulawesi: Phalaenopsis venosa Shim & Fowlie

Phalaenopsis venosa Shim & Fowlie adalah anggrek endemik Sulawesi terancam punah sehingga perlu dilakukan upaya konservasi. Tujuan penelitian untuk mengetahui mikromorfologi dan media perkecambahan in vitro mendukung konservasi P. venosa. Biji dipanen pada enam bulan setelah penyerbukan disimpan di freezer suhu -4 °C selama sembilan bulan. Pengamatan menggunakan mikroskop cahaya OPTIKA M-699. memiliki panjang 91.99±17.73 µm, lebar 20.64 ±4.34 µm; serta embrio dengan 52.27±12.13 16.72±3.42 rongga udara 13.25% ± 11.65%. Pengujian rancangan acak lengkap satu faktor, yaitu macam media, ulangan. Media yang diujikan Hyponex modifikasi (HS), Vacin Went (mVW), Knudson’s C (mKC), (KCA), Murashige Skoog setengah konsentrasi (1/2MS), (MS). Tingkat tertinggi 12 minggu tanam (12 MST) terdapat HS (70.52%) rata-rata 14.35 biji berkecambah. Perkembangan protokorm lebih lambat dibandingkan perkembangan 1/2MS menunjukkan pembentukan rhizoid pucuk daun. Hal itu karena kaya nutrisi HS.
